According to whole-genome sequencing, Aspergillus niger produces multiple enzymes of glycoside hydrolases (GH) 31. Here we focus on a GH31 α-glucosidase, AgdB, from A. niger . AgdB has also previously been reported as being expressed in the yeast species, Pichia pastoris ; while the recombinant enzyme (rAgdB) has been shown to catalyze tranglycosylation via a complex mechanism. We constructed an expression system for A. niger AgdB using Aspergillus nidulans . To better elucidate the complicated mechanism employed by AgdB for transglucosylation, we also established a method to quantify glucosidic linkages in the transglucosylation products using 2D NMR spectroscopy. Results from the enzyme activity analysis indicated that the optimum temperature was 65 °C and optimum pH range was 6.0–7.0. Further, the NMR results showed that when maltose or maltopentaose served as the substrate, α-1,2-, α-1,3-, and small amount of α-1,1-β-linked oligosaccharides are present throughout the transglucosylation products of AgdB. These results suggest that AgdB is an α-glucosidase that serves as a transglucosylase capable of effectively producing oligosaccharides with α-1,2-, α-1,3-glucosidic linkages. 相似文献

Even a zero-power reactor core containing highly enriched uranium has a weak neutron source inherent in uranium 235, and consequently, a neutron counter placed closely to the core without external neutron source registers a certain counting rate. The study of the counting is very important for zero-power reactor physics experiments with a high precision. In this experimental study, first, at a shutdown state of the UTR-Kinki reactor without start-up neutron source, a pulse height distribution of output signals from a neutron proportional counter was measured to confirm that these signals resulted from neutron detections. At several subcritical states of the UTR, then, the Feynman-α analysis was carried out to confirm that the neutrons detected by the counter must be fission neutrons multiplied by fission chain reactions. The correlation amplitude measured in the Feynman-α analysis was much higher than that measured in a previous drive by start-up source. Further, it was also confirmed that the subcriticality dependence of neutron counting rate followed the source multiplication formula. This feature indicated that the one-point model was very successful in the subcritical range including the shutdown state. 相似文献

A prototype ultrasonic liquid film sensor was applied to a high-temperature steam–water two-phase flow experiment. The liquid film sensor was vertically installed in a loop which was connected to HUSTLE, a multi-purpose steam source test facility. The hydraulic diameter of the measurement section was 9.4 mm. The output waveforms of the sensor were acquired with a digital oscilloscope. The fluid temperature and system pressure were kept at 288 °C and 7.2 MPa, respectively, during the experiment. The pulse-echo method was used to calculate the liquid film thickness. The cross-correlation calculation was utilized to determine the time difference between the pulse reflected at the sensor surface and the pulse reflected at the liquid film surface. The time-averaged liquid film thicknesses were less than 0.055 mm in the annular flow condition. The increase of the time-averaged thickness was small with the change of the gas momentum flux. The film thicknesses measured with the sensor were compared with the past experimental results; the former were smaller than one-fourth of the thickness estimated as the mean film thickness. The comparison results suggested that the continuous liquid sublayer thickness was measured with the liquid film sensor. 相似文献

Gamma-ray emission probabilities of 241, 243Am and 239Np have been precisely measured with gamma- and alpha-ray spectroscopic methods. The activities of the samples were determined by measuring alpha particles using a Si semiconductor detector. Gamma rays emitted from the samples were measured with a planar type High-Purity Germanium (HPGe) detector. An efficiency curve of the HPGe detector was derived with uncertainties from 0.7% to 2.5% by combining measured efficiencies and Monte Carlo simulation. The gamma-ray emission probabilities for the major gamma rays of these nuclides were determined with uncertainties less than 1.2%. 相似文献

promotes the crystallization in the skin layer but contributes to increasing the packing of amorphous chains near the skin,

Abstract We propose an approach to manipulate the convergence in multiple solutions of phase conjugate reflectivity in photorefractive four-wave mixing. Although a method forcibly adding a π-phase shift to an incident beam has been already proposed to control the reflectivity, some restrictions have been required in the boundary conditions for the successful operation. Here, we control the reflectivity with the boundary conditions in which the phase shift operation is ineffective by itself. In our method, the phase shift operation is combined with the procedure of turning an incident beam on and off. With a numerical analysis of four-wave mixing, we show that our new approach brings drastic change in the spatial distribution of the index grating and leads the phase conjugate reflectivity which was not manipulated previously. 相似文献

Many research groups are developing series voltage compensators. In a series converter, since a transformer is used in series in the power system, the power system current flows into the voltage source inverter through the transformer. The inverter current, which is determined by the transformation ratio, gives rise to an error voltage that consists of a dead‐time voltage and an impedance voltage. The error voltage is generated even when the reference voltage is zero. The renewable energy source like wind energy generates electric power with intermittent nature. Hydrogen energy system can help to solve the fluctuation problem of the wind power. Totalized Hydrogen Energy Utilization System (THEUS) consists of a Unitized Reversible Fuel Cell (URFC), a hydrogen storage tank, and other auxiliary components. Wind power is inherently variable; the URFC will be subjected to a dynamic input power profile in water electrolyzer mode operation. This paper describes the THEUS operation and performance at different variations in intermittent wind power. The performance of the THEUS was evaluated in water electrolyzer and fuel cell mode operation. The stack efficiency, system efficiency, and system efficiency including heat output from the URFC were presented at each operation. The total efficiency of the URFC and THEUS were also investigated. The maximum total efficiency of the URFC and THEUS were 53% and 66%, respectively. 相似文献

The phase equilibrium temperature and dissociation heat of tetrabutylammonium propionate (TBAPr) hydrate are reported. TBAPr hydrate is a type of ionic semiclathrate hydrates and also could potentially be used as thermal energy storage material. The temperature‐composition phase diagram of the TBAPr hydrate was determined in a defined range of mass fractions. Considering the dissociation heat of differential scanning calorimetry (DSC) measurements, multiple peaks of heat flow were observed in the TBAPr‐water system at the TBAPr mass fraction lower than 0.35, and there was a single peak at the mass fraction higher than 0.37. 相似文献
